脑出血大鼠心肌组织炎性因子及热休克蛋白表达的变化及扶正护脑胶囊的干预作用

摘  要:目的观察脑出血大鼠心肌组织炎性因子及热休克蛋白表达的变化及扶正护脑胶囊的干预作用。方法动物随机分为假手术组、模型组、扶正护脑组和安宫牛黄组,分别于术后6、24、72h3个时相点取材,采用免疫组织化学法检测肿瘤坏死因子-α(TNF-α)、热休克蛋白70(HSP70)的蛋白表达,通过图像分析,测定IOD值。结果模型组各时间点TNF-α、HSP70的IOD值均显著升高,扶正护脑组和安宫牛黄组均能明显降低TNF-α的IOD值,升高HSP70的IOD值。结论炎性因子参与了脑出血诱发心肌损伤的机制,扶正护脑胶囊通过抑制炎性因子表达、增强热休克蛋白表达而减轻心肌损伤。Objective To observe the effect of Fuzheng Hunao capsule on TNF-a and HSP70 protein expression of myocardial tissues in rats with intracerebral hemorrhage. Methods Seventy two rats were randomly divided into 4 groups: sham, model, Fuzheng Hunao and Angong Niuhuang. The protein expression of TNF-a and HSP70 in rat myocardial tissues were detected by the immunohistochemistiy method, and the integrated optical density (IOD) was measured with image analysis. Result The IOD of TNF-α and HSP70 in model group was higher than that in sham group. The IOD of TNF-α in Fuzheng Hunao and Angong Niuhuang group was lower than that in model group. While the IOD of HSP70 was higher than that in model group. Conclusion Inflammatory response attend the myocardial damage. Fuzheng Hunao capsule could protect the myocardial tissues by inhibiting the protein expression of TNF-a and increasing that of HSP70.
