摘  要:目的观察人参皂苷Rg1、Rb1对淀粉样前体蛋白代谢途径中α分泌酶活性和基因表达的影响。方法用转染突变型PS1(ML46L)和野生型PS1的CHO细胞,通过荧光酶标技术检测α分泌酶活性,Western blot免疫印迹、RT-PCR技术检测α分泌酶基因ADAM9、ADAM10的蛋白和RNA表达。结果转染突变型PS1 ML46L组α分泌酶活性较转染野生型PS1(WT组)低(P<0.05),加用人参皂苷Rg1、Rb1后转染突变型PS1ML46L组α分泌酶活性较未加用组不同程度增高(P<0.05),同时ADAM9、ADAM10的蛋白及RNA表达较未加用组有不同程度增多(P<0.05)。结论人参皂苷Rg1、Rb1均可不同程度增加α分泌酶中ADAM9、ADAM10基因表达,提高α分泌酶活性,且人参皂苷Rg1调节作用优于人参皂苷Rb1。关键词:人参皂苷Rg1;人参皂苷Rb1;淀粉样前体蛋白分泌酶;Objective To study effects of Ginsenoside Rgl, Rbl on activity and genetic expression of alpha secretase in the progress of metabolism of APP. Methods Chinese hamster ovary (CHO) cell line transacted by mutant PS 1 (MIA6L) genes and wild type PS 1 genes was used. The activity of alpha secretase was tested by fluorometric reaction. The expression of ADAM9 and ADAM10 was analysis through western blotting and RT-PCR. Results The activity of alpha secretase was increased significantly in Ginsenoside Rgl, Rbl group than that without Ginsenoside group (P〈0.05). The protein and RNA expression of ADAM9 and ADAM10 treated with Ginsenoside was higher than that untreated (P〈0.05). Conclusion Both Ginsenoside Rgl and Rbl could increase the protein and RNA expression of ADAM9 or ADAM 10, upregulate the activity of alpha secretase. Ginsenoside Rgl is more effective than Ginsenoside Rbl.
