摘 要:为比较固定化金属亲和层析膜与亲和层析胶的纯化效果,分别用固定化金属亲和层析胶Cu2+柱、Ni2+柱、固定化金属亲和层析膜Cu2+柱、Ni2+柱分离纯化rhCu,Zn-SOD、rhMn-SOD/24a、rhMn-SOD/28。结果表明,亲和胶与亲和膜分离SOD获得相似的纯化效果,纯化倍数均在3到5之间,亲和膜分离所得SOD比活略高于亲和胶,rhCu,Zn-SOD与Cu2+的亲和能力高于rhMn-SOD。固定化金属亲和层析膜色谱为基因工程产物的快速分离鉴定提供了一个简便的方法。The purpose is to compare the resolution effect of agarose immobilized metal affinity chroma- tography and membrane immobilized metal affinity chromatography. IMAC agarose and membrane charged with Cu^2+ ion and Ni^2+ ion were used to purify rhCu,Zn-SOD, rhMn-SOD/24a and rhMn-SOD/ 28a respectively. The results suggested that IMAC agarose and IMAC membrane had the similar effect in purification of SOD. The purified factor is between 3 and 5. The specific acitivity of SOD purified through IMAC membrane was a little higher than that of IMAC agarose. The affinity to Cu^2+ ion of rhCu,Zn-SOD was stronger than that of rhMn-SOD. IMAC membrane was a new, simple method for purification of gene engineering products.
