摘  要:目的比较新分离的新城疫病毒野生株(NDV7793、NDVD817)与疫苗株Lasota对大肠癌的体外杀伤作用。方法3株病毒作用于大肠癌细胞株LS174T和LOVO,用乳酸脱氢酶微量释放法测定病毒的杀瘤活性并通过测定培养上清液的血凝效价来检测病毒的增殖能力。结果3株新城疫病毒均可杀伤大肠癌细胞并可在其复制增殖,对正常大肠细胞无明显细胞毒性。而对正常大肠组织细胞未见明显影响(P<0.05);野生株对癌细胞的杀伤作用较强,72h后对LOVO细胞株的杀伤效应>90%;LOVO大肠癌细胞对NDV敏感性强于LS174T大肠癌细胞。结论3株NDV对大肠癌细胞均有选择性杀伤作用,野生株病毒的抑瘤效应强于疫苗株,对正常大肠组织无细胞毒性。Objective To study the antitumor activity of human colorectal cancer by Newcastle Disease Virus wild strains( NDV 7793,NDV D817) and NDV vaccine strain in vitro. Methods Three NDV strains active on human h colorectal cancer cell lines LS174T and LOVO,The tumor-cytotoxic effects of NDV were detected by LDH release assay and the reproductive activities of NDV were observed by haemagglutination (HA) titre. In the end,tumor-cytotoxic effects of NDV wild strains and NDV vaccine were furtherly compared. Results The apparently pathological changes and the reproductive activities of wild NDV strains were observed in LS174T and LOVO tumor cells, but not in normal colorectal cells. The higher suppressed effect on LOVO tumor cells than on LS174T tumor cells in three NDV strains. Conclusion Wild NDV strains can be a potential anticancer agent for the treatment of colarectal cancer.
