HPLC法测定不同市售荷叶药材中4种生物碱类成分的含量

摘　　要：目的建立测定荷叶药材中4种生物碱类成分2-羟基-1-甲氧基阿朴啡、原荷叶碱、荷叶碱和莲碱含量的高效液相色谱方法,并测定不同市售荷叶药材中4种成分的含量。方法采用HypersilC18柱（250 mm×4.6 mm,5μm）,以乙腈-0.1%三乙胺水溶液为流动相,梯度洗脱,流速1.0 mL/min,柱温35℃,检测波长270 nm。结果2-羟基-1-甲氧基阿朴啡、原荷叶碱、荷叶碱和莲碱的线性范围分别为0.1096-0.6576μg（r=0.9995）,0.0210-0.1260μg（r=0.9995）,0.1030-0.6180μg（r=0.9998）,0.0856-0.5136μg（r=0.9995）,平均加样回收率（n=6）分别为99.5%,100.5%,97.5%和100.1%。30批市售品中4种生物碱类成分的含量分别为：2-羟基-1-甲氧基阿朴啡0.010%-0.076%,原荷叶碱0.002%-0.120%,荷叶碱0.008%-0.249%,莲碱0.008%-0.067%。结论30批市售荷叶药材中4种生物碱类成分含量变化较大,其中原荷叶碱和荷叶碱的含量变化更为明显,应将4种生物碱同时纳入荷叶药材的质量控制。Objective To establish a HPLC method for the quantitative determination of four alkaloids,i.e.2-hydroxy-1-methoxyaporphine,pronuciferine,nuciferine and roemerine,in Nelumbo nucifera.Methods The determination was carried out with Hypersil C18 column（250 mm × 4.6 mm,5 μm）,using acetonitrile-water（0.1% triethylamine） as mobile phase with gradient elution,at a flow rate of 1.0 mL·min-1 and detected at the wavelength 270 nm,column temperature 35 ℃.Results The linear ranges of 2-hydroxy-1-methoxyaporphine,pronuciferine,nuciferine and roemerine were 0.109 6～0.657 6 μg（r=0.999 5）,0.021 0～0.126 0 μg（r=0.999 5）,0.103 0～0.618 0 μg（r=0.999 8） and 0.085 6～0.513 6 μg（r=0.999 5）,with the average recoveries（n=6） of 99.5%,100.5%,97.5% and 100.1% respectively.The contents of four components in different samples of Nelumbo nucifera were： 2-hydroxy-1-methoxyaporphine 0.01%~0.076%,pronuciferine 0.002%~0.120%,nuciferine 0.008%~0.249%,roemerine 0.008%~0.067.Conclusion The contents of 4 components in 30 market samples varied notably,especially for pronuciferine and nuciferine,and should be taken into the quality control of Nelumbo nucifera.
