摘 要:目的:建立肾间质纤维化大鼠模型,动态观察模型大鼠肾功能、肾组织中HGF mRNA的表达变化及补肾活血方对其影响,探讨补肾活血方治疗慢性肾功能衰竭的可能作用机制。方法:将大鼠随机分为假手术组、模型组、洛汀新组、补肾活血方高剂量组、补肾活血方低剂量组,除假手术组外其余大鼠采用单侧输尿管梗阻的方法建立肾间质纤维化大鼠模型;分别于UUO术后3、7、14、21、28d观察造模大鼠血肌酐、尿素氮的动态变化;采用RT-PCR法,观察造模大鼠肾组织中肝细胞生长因子(HGF)mRNA的表达变化及补肾活血方对其影响。结果:与假手术组比较,造模大鼠血清肌酐、尿素氮浓度呈进行性增高,P<0.05或P<0.01;HGF mRNA表达于UUO术后第3天开始增加,于第14天达高峰,P<0.05或P<0.01,随后随时间延长表达逐渐渐弱。与模型组比较,各给药组大鼠肾功能改善,P<0.05或P<0.01;各时间点HGFmRNA的表达呈不同程度增加,P<0.05或P<0.01。结论:补肾活血方可以改善肾间质纤维化大鼠肾功能;可以提高肾组织中HGFmRNA的表达,防治肾间质纤维化。Objective: to found renal interstitial fibrosis rat model and observe the HGFmRNA express change in model rats nephridial tissue, renal function and the effect of the prescription for invigorating the kidney and promoting blood flow and to discuss the mechanisrn of the prescription for invigorating the kidney and promoting blood flow to treat chronic renal failure. Method: the rats were randomly divided into the sham operated, model, Lotensin, the high dose prescription for invigorating the kidney and promoting blood flow and the low dose prescription for invigorating the kidney and promoting blood flow group. The method of half obstruction of ureter was used in the model of renal interstitial fibrosis except the sham operated. The rats serum creatinine and urea nitrogen were observed in 3 days, 7 days, 14 days, 21 days and 28 days after UUO. The express change of HGFmRNA in nephridial tissue and the effect of the prescription for invigorating the kidney and promoting blood flow were observed. Result: the serum creatinine and urea nitrogen was increase in the model than sham operated and the HGFmRNA express was increased after 3 days and peak appeared in the 14 days. After that, the HGFmRNA express was tapered. The drugs rat's renal function was amendment and the HG-FmRNA express was increase. Conclusion: the prescription for invigorating the kidney and promoting blood flow could improve renal interstitial fibrosis rat's renal function and improve nephridial tissue HGFmRNA express and prevention and cure renal interstitial fi- brosis.
