摘  要:目的观察超滤膜提取当归补血汤对荷瘤小鼠脾细胞白细胞介素-2(IL-2)和γ-干扰素(IFN-γ)的诱生作用,探讨其对荷瘤小鼠免疫系统的调节机制。方法采用超滤膜分离技术提取当归补血汤,用双抗体夹心ELISA法检测培养脾淋巴细胞上清液中IL-2和IFN-γ的含量,用反转录-聚合酶链式反应(RT-PCR)法测定脾淋巴细胞IL-2和IFN-γmRNA的表达水平。结果超滤膜提取当归补血汤各剂量组小鼠脾淋巴细胞培养上清液中IL-2和IFN-γ的含量均较模型组增高,大剂量组与模型组比较有显著性差异(P<0.05)。各剂量组小鼠脾淋巴细胞IL-2和IFN-γmRNA的表达水平均增高,中剂量组和大剂量组与模型组比较,均有显著性差异(P<0.05)。结论超滤膜提取当归补血汤能够促进荷瘤小鼠脾淋巴细胞细胞因子IL-2和IFN-γ的分泌及其mRNA的表达,从而发挥免疫调节作用。Objective To evaluate the effect of ultra-filtration extract from Danggui Buxue Decoction (DBD) on immune function of H22 bearing mouse and explore the mechanism through observing the effect of ultra-filtration extract from DBD on the secretion of the cytokines (IL-2, IFN-γ). Methods We used membrane separation technique to extract DBD. The content of IL-2 and IFN-γ in splenocyte culture supernatant was detected by enzyme-linked immunosorbent assay (ELISA), and the mRNA expression of cytokines (IL-2, IFN-γ ) in splenocytes was assayed by reverse transcription-polymerase chain reaction (RT-PCR). Results The content of IL-2 and IFN- γ in splenocyte culture supernatant was increased in ultra-filtration extract from DBD groups, significantly in high-dose group (P〈0.05). The mRNA expression of IL-2 and IFN- γ was higher in DBD groups than that in negative control, markedly higher in medium-dose group and high-dose group (P〈0.05). Conclusion Ultra-filtration extract from DBD groups has the function of facilitating the secretion and gene transcription of cytokines (IL-2, IFN- γ ) in H22 bearing mouse. It can improve the immune function of tumor-bearing mouse and inhibit the growth of the tumor.
