摘  要:[目的]为发状念珠藻多糖的提取、开发及生物学活性研究提供试验及理论基础。[方法]以发状念珠藻为材料,用石油醚脱脂脱色素,80%的乙醇去可溶性小分子糖类,用水作提取剂,98℃下提取3次,每次3h,合并提取液后减压浓缩,三氯乙酸脱蛋白,SephadexG-50层析纯化,醇析后冷冻干燥,进行红外光谱测定和种子发芽试验。[结果]结果表明,发状念珠藻多糖得率为15.8%~19.7%,红外光谱的特征吸收峰明显,是一种含氨基和B糖苷键,不含硫酸基的多糖。当多糖浓度为1.0mg/L时油菜种子、小麦种子、水稻种子和胡麻种子的发芽率分别比对照组增加14.29%、12.79%、4.40%和15.48%,而多糖浓度为2.0mg/L时白菜和玉米种子的发芽率分别比对一照组增加11.24%和22.50%。[ Objective ] The research aimed to provide experimental and theoretical basis for the extraction and biological activities of polysac- charides from Nostocflagelliforme. [ Method ] Taking N. flagelliforme as the material, after refluxing with petroleum aether to remove fatty materials and pigments, 80% alcohol was used to remove dissolubility saccharide with lower molecular weight, extracting for 3 times with water at 98 ℃ for 3 h. The extracting solutions were concentrated under vacuum after blending raffinate, After protein removal by TCA, Sephadex G-50 chromatography purification was carried out, Seed germination test was conducted by infrared spectrum, [ Result] The results showed that the yield of polysaccharide was 15.8% - 19.7%. Characteristic absorption 'peak of infrared spectrum was obvious. The substance was a kind of polysaccharide with -NH2 and IB-glycosidic linkage, but without -SO4. The average germination percentage of Brassica chinensis, Semen tritici aestivi, Oryza satival and Linum usitutissimum increased by 14.29 %, 12.79%, 4.40% and 15.48% , respectively, compared with the control when added 1.0 mg/L polysaccharides of N. flagelliforrne. While polysaccharides concentration being 2.0 mg/L, the germination rates of Brasslca campestris and Zea rnays enhanced by 11.24% and 22.50% , respectively.
