摘  要:目的建立黄芩RP-HPLC指纹图谱,考察黄芩不同生长发育期的主要化学成分的变化。方法色谱柱为Diamonsil C18(4.6mm×250mm,5μm),以乙腈-水-甲酸(21∶78∶1,A)和乙腈-水-甲酸(80∶20∶1,B)为流动相进行梯度洗脱;检测波长280nm;柱温30℃;流速1.0mL·min-1。对黄芩指纹图谱进行分析,并比较了10个化学成分含量在不同生长发育期的变化趋势。结果黄芩中主要化学成分在不同生长发育期的积累具有一定规律。苷类和苷元类成分含量在生长过程中虽发生了不同的变化,但最终二者含量均在枯黄期恢复到接近休眠期时的含量水平。结论揭示黄芩中主要化学成分在不同生长发育期的变化趋势。OBJECTIVE To develop a RP-HPLC method of fingerprint analysis for ScuteUaria baicalensis, and to investigate the variation of main components in samples at different growthperiods. METHODS Analysis were carried out on a ODS Diamonsil( C18, 4.6 mm ×250 mm,5 μm)column at 30 ℃. The mobile phase was in the gradient elution at a flow rate of 1, 0 mL · min^-1. Mobile phase A consisted of acetonitrile, water and formic acid (21: 78: 1 )while mobile phase B consisted of acetonitrile, water and formic acid( 80: 20: 1 ). The detection wavelength was set at 280 nm. The fingerprints of ScuteUaria baicalensis were analyzed, and the content change trend of 10 ingredients in samples at different growth was compared. RESULTS There were some rules for the change of main components in ScuteUaria baicalensis at different growth periods. Although the contents of flavonoid glycosides and flavonoid aglycones varied during the development, at last both recovered approximately to the content level of dormancy period at yellow period. CONCLUSION The variation trend of main components in ScuteUaria baicalensis at different growth periodswas found.
