摘  要:目的:建立测定人体内羟苯磺酸钙血浆药物浓度的高效液相色谱法。方法:Diamonsil C_(18)色谱柱(200 mm×4.6 mm,5μm);流动相:乙腈-10 mmol·L^(-1)醋酸铵(含0.3%三乙胺,用冰醋酸调节 pH 为3.5)(7:93),流速为1.0 mL·min^(-1),柱温为40℃,检测波长305 nm。结果:羟苯磺酸钙在0.36~22.78μg·mL^(-1)(r^2=0.9998)范围线性关系良好,最低定量限为0.36μg·mL^(-1),提取回收率大于50%,日内和日间 RSD 均小于10%。结论:此方法能够简便、高效、快速和灵敏地检测健康志愿者血浆中的羟苯磺酸钙,可用于羟苯磺酸钙药物代谢动力学研究。Objective:To establish an HPLC method for determination of calcium dobesilate in human plasma. Methods:Chromatographic column was Diamonsil C 18 (200 mm × 4.6 mm ,5 μm) ;the mobile phase were composed of acetonitrile- 10 mmol·L^-1 ammonium acetate (0. 5% triethylamine and pH was regulated to 5.5 with glacial acetic acid) (7: 95) ,at the flow rate of 1.0 mL · min^-1 ,the column temperature was 40 ℃ and the detector wave- length was set at 305 nm. Results:The calibration curve was linear in the range of 0. 36 -22.78 μg· mL^-1(r^2 = 0. 9999) for determination of calcium dobesilate. The extract recovery was more than 50%. The RSDs of inter - day and intraday were all less than 10%. Conclusions:The method is simple, rapid, high efficient,sensitive and suitable for determination calcium dobesilate in healthy volunteers' plasma, it can be used in pharmacokinetics and bioequivalence research of calcium dobesilate.
