摘  要:目的探讨庆大霉素对耳蜗内毛细胞带状突触数量的影响及其意义。方法C57小鼠每日庆大霉素(100mg/kg)进行腹腔注射,分别于第4d、第7d、第10d、第14d取耳蜗基底膜顶回作为实验组(每组5只);取正常小鼠(5只)耳蜗基底膜顶回作为对照组。使用免疫荧光双标法对突触前膜RIBEYE和突触后膜GluR2&3进行标记,激光共聚焦显微镜进行光学连续切片,3DMAX进行三维建模,对内毛细胞带状突触进行计数。结果所有小鼠内毛细胞均无缺失,但内毛细胞带状突触数量存在差异(P<0.01)。带状突触数量在庆大霉素腹腔注射第4天接近正常,第7天时明显高于正常,10天时接近正常,14天时明显低于正常。结论庆大霉素作用14天,内毛细胞带状突触数量明显低于正常,可能是损伤听力的机制之一。Objective To investigate the change of the ribbon synapses in inner hair cells by using gentamicin. Methods C57 mice were intraperitoneally injected with gentamycin(100mg/kg) every day, whose apical coil of organ Corti were obtained at 4th day, 7th day, lOth day and 14th day respectively in experimental group (every group, n =6), and 6 mice as normal control (n =6). We marked the presynaptic membrane with RIBEYE and the postsynaptic membrane with GluR2& 3 by double labeling of immunofluorescence, successively sliced by laser confocal microscopy, three dimension restructed by 3ds max 8, and then counted the ribbon synapses in inner hair cells. Results The inner hair cells of all mice were intact, but there were different numbers of the ribbon synapses in inner hair cells between groups(P〈0.05). The number of the ribbon synapses in inner hair cells at 4th day and 10th day of gentamicin injection was similar to normal, but higher for 7th day of gentamicin injection than in normal, however, much lower for 14th day of gentamicin injection than in normal. Conclusion The loss of the ribbon synapses in inner hair cells by gentamicin for 14days may be one of the mechanisms for the damage of hearing ability.
