摘 要:在三氯乙酸酸性底液中,苯甲醛在过量的条件下与异烟肼反应生成异烟肼-苯甲醛腙。反应后溶液再加入NaOH溶液调节酸度,用极谱法测定异烟肼-苯甲醛腙,发现在-0.848V(vs.SCE)处产生一灵敏的二阶导数极谱还原波,比相应的异烟肼极谱还原波灵敏度提高了50倍以上。基于此,拟定了测定血清中异烟肼的新方法。本法测定的异烟肼在1.0×10^-4~3.5×10^-2mg/L范围内呈良好的线性关系,相关系数r^2=0.9989。检出限可达2×10^-5mg/L,方法可用于血清中异烟肼的测定。Derivative polarographic analysis is developed for the determination of isoniazid (INH) in serum. It is based on the formation of the hydrazone with isoniazid and benzaldehyde in the presence of trichloroacetic acid. The sensitive second derivative polarographic wave of isoniazid-benzaldehyde hydrazone is obtained at - 0. 848 V ( vs. SCE). A sensitivity of the hydrazone is 50 times larger than that of isoniazid under optimum conditions and the peak current is propertional to the concentration of isoniazid over the range from 1.0 × 10^-4 mg/L to 3.5 ×10^-2 mg/L ( r^2 = 0.9989) with limit of detection of 2 × 10^-5 mg/L. This method has potential to be used for analysis of isoniazid in serum.
