摘  要:目的:探讨血管紧张素转化酶(ACE)基因插入/缺失多态性及活性与Ⅱ型糖尿病肾病的关系。方法:应用聚合酶联反应方法检测157例Ⅱ型糖尿病患者和50例健康对照的ACE基因型,同时检测血浆ACE水平。结果:糖尿病肾病组的DD基因型频率及D等位基因频率都高于对照组的相应频率,差异有统计学意义(P<0.05)。DD基因型ACE水平高于II基因型。结论:ACE基因的DD基因型与血浆ACE活性有关,ACE基因缺失多态性可能为Ⅱ型糖尿病肾病的遗传因素。Objective: To investigate the relationship among the ACE activity, ACE gene I/D polymorphism and diabetic nephropathy, Methods: 157 patients meeting the criteria of type 2 diabetic mellitus were recruited,including 65 patients with overt nephropathy. The control group includes 50 age-match health cases. The I/D polymorphism of ACE gene was analyzed by PCR amplification. The serum ACE activity of all the patients and control group were measured simultaneously. Results: The frequency of the ACE DD genotype in the patients with diabetic nephropathy was significantly higher than in the control group. The serum ACE activity was higher in DD homozygotes than in Ⅱ homozygotes. Conclusion: The DD genotype of ACE gene attributes to the susceptibility of diabetic nephropathy. The detection of ACE activity is useful for the diagnosis and therapy.
