摘  要:目的探讨表皮葡萄球菌ica操纵子与细菌间多糖粘附素(polysaccharide intercellul aradhesin,PIA)及生物膜表型之间的关系。方法采用聚合酶链反应(PCR)检测icaA基因,刚果红琼脂平皿检测PIA,微量板半定量法检测细菌生物膜表型;统计学分析icaA基因与PIA及生物膜表型之间的相关性。结果49株临床分离菌株中,32.7%(16/49)能形成生物膜,67.3%(33/49)无生物膜形成。icaA基因阳性菌株中80%(8/10)有生物膜形成,ica操纵子与生物膜表型密切相关(P<0.01);PIA阳性菌株中63.6%(14/22)有生物膜形成,PIA与生物膜表型密切相关(P<0.01)。结论表皮葡萄球菌ica操纵子的存在与生物膜表型密切相关,PIA的合成是生物膜形成的关键环节。Objective To investigate the relationship among ica operon of Staphylococcus epidermidis (SE), polysaccharide intercellular adhesion (PIA) and biofilm phenotype.Methods ica operon was detected by PCR to amplify icaA gene. Congo red agar plate was used to examine PIA synthesis. Semi-quantitative test was performed to examine biofilm phenotype. The statistical significance of the relationship among icaA, PIA and biofilm phenotype was evaluated. Results Among 49 selected SE strains, biofilm formation was positive in 16 (32.7%) strains while negative in 33 (67.3%) strains. 80% of icaA-positive strains presented biofilm formation, indicating a close relationship between ica operon and biofilm phenotype (P〈0.01). 63.6% of PIA-positive strains showed biofilm formation, implying a close relationship between PIA and biofilm phenotype (P〈 0.01). Conclusion The presence of ica operon is statistically associated with biofilm formation. PIA synthesis is the key link of biofilm formation.
