摘 要:目的:通过观察艾炷灸"足三里、悬钟"两穴对衰老小鼠模型的学习记忆功能及脑组织NO含量、NOS活力的影响,探讨艾炷灸对脑老化的作用机理。方法:36只小鼠随机分为3组,将两组连续注射D-半乳糖6周,其中一组模型组,另一组为在12天后进行艾炷灸治疗(治疗组),第3组注射同剂量生理盐水为对照组,另将16月龄小鼠作为第4组自然衰老对照组(自衰组)。采用Morris水迷宫检测小鼠学习记忆功能,NO含量、NOS活力测定分别用硝酸还原酶法和化学比色法。结果:(1)在水迷宫测试的6项指标中①模型组与对照组比较"潜伏期、寻求次数、停留时间、象比"4项指标均有显著性差别(P<0.05),模型组与自衰老组在"寻求次数、停留时间、跨越次数、原平台象比"4项均无差别(P>0.05),提示模型组的学习记忆能力与自衰组相似。②模型组与治疗组比较6项均有显著性差异(P<0.05,P<0.01),治疗组与对照组各项指标无显著性差异(P>0.05)。(2)①模型组和自衰组的大小脑皮质NO含量、NOS活力都比对照组明显升高(P<0.05,P<0.01),提示D-半乳糖造成亚急性衰老小鼠大小脑皮质NO含量、NOS活力都明显升高,与自然衰老小鼠的情况接近。②模型组与治疗组比较,大小脑皮质NO含量、NOS活力显著升高,有显著差异(P<0.05,P<0.01);对照组与治疗组比较,无显著差异(P>0.05)。结论:艾炷灸"足三里、悬钟"两穴可以改善D-半乳糖所致衰老小鼠的学习记忆功能,与其可降低异常增高脑组织中NO含量、NOS活力有关,具有延缓衰老作用。Objective: To explore the effects of moxa cone moxibustion of "zusnali" (ST36)and "xuanzhong" (GB39)on learning and memory function and Nitric Oxide(NO) and Nitric Oxide Synthase(NOS) of Cerebral Tissues in D-galactose induced Aging Mice. Methods : After selecting with the Morris water maze(MWM) test, 36 female mice(3 months old) were randomly and equally divided into the normal sodium control group, the model group and the treatment group. In addition, 12 female mice(16 months old) were selected as the physio-senility control group. The treatment group and the model group were induced by D-galactose injection for 42days,while the normal sodium control group was injected the same dosage of normal sodium for 42 days. From the 13th day, the treatment group was received moxa cone moxibustion therapy for every other day until the model would had been finished. At the end of treatment, the four groups were tested on Spatial Learning and Memory in the MWM again, and then examined the NO contents and the NOS activities of cerebral tissuesatlesults : Thirty-six female mice were equal on spatial learning and memory before the experiment(P〉0.05), after modeled and treated, The significant difference in place navigation test was observed between the model and the normal sodium control group following with the four targets that are escape latency, seeking (platform) frequency, detention (platform) time, percentage of platform quadrant (the percentage of swimming distant between the original platform and total distant) ( P〈0.05 ),there are no differences between the model and the physio-senility control group following with the four targets that are seeking (platform) frequency, detention (platform) time, spanning (original) platform frequency and percentage of (original) platform quadrant(at the 7th day of MWM, after taken away original platform, mice were required to find original place of platform) (P〉0.05).The significant differenc
