摘 要:目的用荧光光谱法研究苯酚和小牛胸腺DNA(calf thymus DNA,ct-DNA)之间的相互作用。方法采用荧光光谱的变化、离子强度的影响、温度的影响等实验研究苯酚与小牛胸腺DNA间的相互作用模式,并根据Stern-Vol mer方程进行数据处理。结果在pH7.0的磷酸盐缓冲溶液中,苯酚的荧光激发峰和发射峰分别位于273nm和299nm处。ct-DNA的加入对苯酚的荧光存在着猝灭作用,室温条件下测定ct-DNA对苯酚的动态猝灭常数为7.062×103L/mol。结论苯酚与ct-DNA之间属于动态猝灭过程,未形成络合物。Objective To investigate the interaction between phenol and calf thymus DNA (ct-DNA). Methods The interaction mode was studied by using the changes of ultraviolet spectrum and fluorescence spectrum, ionic strength effects and temperature effects. The experimental data were plotted by Stern-Volmer equation. Results At pH 7.0 phosphate buffer solution,the excitation and emission wavelength of phenol are 273 nm and 299 nm, respectively. The addition of ct-DNA to phenol solution resulted in fluorescence quenchin^g and Under the room temperature circumstance, the quenching constant Ksv = 7. 062 × 10^3 L/mol. Conclusion The fluorescence quenching between phenol and ct-DNA belongs to dynamic quenching, not Static quenching,which means they don't form new adduct.
