甲状腺切除的肾阳虚大鼠肝线粒体蛋白质组的研究

摘　　要：目的:应用差异蛋白质组学技术研究肾阳虚大鼠肝线粒体蛋白质组。方法:正常大鼠和甲状腺切除的肾阳虚模型大鼠的肝线粒体蛋白质经荧光染料Cy3、Cy5标记,与等量Cy2标记的内标混合后在同一胶中进行电泳分离,经不同光激发后扫描得到不同样品的蛋白质组图谱。经DeCyder6.5软件进行差异蛋白质组分析,并对差异蛋白质进行质谱鉴定。结果:在肾阳虚大鼠肝线粒体中,氨甲酰磷酸合成酶Ⅰ、乳铁传递蛋白、热休克蛋白60、过氧化氢酶、甲基丙二酸半醛脱氢酶、类钙激活中性蛋白酶、电子传递黄素蛋白、酯酶同工酶ES1、谷胱甘肽过氧化物、ATP合成酶和ATP酶质子通道蛋白表达量均增加,而二氢硫辛酰胺支链酰基转移酶、脂酰辅酶A脱氢酶、鸟氨酸转氨酶和鸟苷酸结合调节蛋白表达量降低。结论:肾阳虚大鼠可能因糖类和脂类物质分解代谢受抑以及细胞内信号转导异常而出现虚寒症状。Objective:The technology of proteomics was used to study the Liver mitochondria proteome of the thyroidectomized Kidney-Yang deficiency Rats.Methods:Every liver mitochondria protein sample from the normal rats and the kidney-yang deficiency rats was labeled with Cy3 or Cy5 randomly,Each Cy3-labeled sample and Cy5-labeled sample were mixed on the same 2-D gel along with a Cy2-labeled mixture of all samples as an internal standard and run on the same gel.The gels were scan under different wavelength light after electrophoresis.All images were analyzed by DeCyder image QuantTM 6.5 and the different proteins were identified by mass spectrum.Results:the expression of carbamoyl-phosphate synthetaseⅠ,lactotransferrin,HSP60,Catalase,methylmalonate semialdehyde dehydrogenase,calpain 12,alpha-ETF,Homolog of zebrafish ES1,glutathione peroxidase,ATP synthase and H+-transporting two-sector ATPase increased in the thyroidectomized Kidney-Yang deficiency rats,while the expression of oxoisovalerate dehydrogenase,Acyl-CoA dehydrogenase,Ornithine aminotransferase and GTP-binding regulatory protein decreased.Conclusion:Because the carbohydrate and lipids catabolism were restrained and the signal conduction was abnormal,the kidney-yang deficient rats was shot of energy and appeared the clinic deficiency-cold symptom.
