摘 要:目的:研究青龙衣中胡桃醌的最佳提取方法及该方法的最佳提取工艺。方法:以95%乙醇为溶剂,胡桃醌含量为指标考察冷浸法、热回流法、超声法对青龙衣中胡桃醌含量的影响,确定胡桃醌的最适提取方法。采用L9(34)正交试验法考察各影响因素(A溶剂种类、B溶剂量、C提取时间),以胡桃醌含量为指标进行直观分析及方差分析,确定最佳提取工艺。结果:胡桃醌的最佳提取方法为超声波法,该法的最佳提取工艺条件为:A2B2C3,即氯仿,8倍溶剂量,提取40min,影响胡桃醌提取的主要因素为A(溶剂种类)。结论:此提取工艺简单易行,稳定可行,适于青龙衣中胡桃醌的大规模提取。Objective:To study the optimum extraction method and technology of juglone from Qinglongyi.Methods:Juglone was extracted respectively by ethanol soak,reflux method and ultrasound wave method from Qinglongyi with 95% ethanol.The content of juglone was determined by HPLC,which was taken as the evaluation index to optimize the optimum extraction method.The L9(34) orthogonal table was applied were employed to select the optimum extraction technology by means of solvent species,solvent volumn and extracting time as the three main factors in which all factors been selected of three levels.HPLC method was used to determine the content of juglone in Qinglongyi.Results:The optimum extraction method was ultrasound wave method and the optimum extraction technology were 8 times of chloroform,ultrasound wave and extracting for 1 time and 30 minutes every time.Conclusion:This optimum extradion process of juglone in Qinglongyi is simple and repeatable and it can be used in industrial production.
