摘  要:自2000年起从广西各地采集1563份犬脑、4份发病牛脑、1份发病猪脑和1份猫脑组织。通过对这些动物脑组织进行PCR检测及用小白鼠脑内接种试验分离出42株狂犬病毒株,并对N基因3’端455个核苷酸片段进行测序分析。根据核苷酸同源性及遗传进化树分析结果,广西狂犬病流行毒株可分成4个群。Ⅰ群有GX01、GX08、GX09、GX014、GX019、GX091、GX123、GX173、GX174、GX195、GX260、GX443、GX452、GXSIO、GX520、GXBS、GXSL、GXGG、GXLA、GXHX、GXGL、GXNniu、GXWXp共23株,它们的核苷酸同源性在97.6%-100%之间;Ⅱ群有Gx074、Gx219、GX304、GX441、GX442、GX496、GX0510、GXBX、GXBM、CXQZ共10株,它们的核苷酸同源性在98.5%-100%之间;Ⅲ群只有GXN119;Ⅳ群有G)(506、GXS08、GXS09、GX120、GXB03、GXeat、GX201、GX102共8株,它们的核苷酸同源性在99.1%-99.6%之间。进一步氨基酸变异分析得知,与兽用疫苗株E1LA比较,所有42个毒株的第58位氨基酸由P变为S,Ⅰ、Ⅱ、Ⅲ群的34株共同发生了H26Y、C40S、S61N、V95L、G106D和P135S位点的变异,Ⅰ群发生了T90N和E110D、Ⅱ群发生了T42S的群特异性变异。氨基酸分析结果与核苷酸同源性、遗传进化树分析结果一致。各群毒株呈现明显的地域分布特征,Ⅰ群主要分布在桂东地区,Ⅱ群主要分布在桂西地区。Ⅳ群主要分布在桂南地区。In this study,1569 samples containing 1563 dogs, brains,4 cattle's brains,1 pig's brain and 1 cat' s brain,were collected from different places of Guangxi since 2000.42 samples were determined to be the rabies positive using reverse transcription polymerase chain reaction (RT-PCR) and the rabies vires was isolated by mouse inoculation test. The length of 455 nuclcotides of 3' terminal N gene of 42 rabies virus isolates were amplified by RT - PCR,then the fragments of N genes were cloned and sequenced. The homology of nucleotide and phylogenetic tree analysis showed that all rabies vires isolates from Guangxi belong to genotype Ⅰ and could be divided into 4 groups as Ⅰ , Ⅱ, Ⅲ and Ⅳ. Group Ⅰ includes 23 isolates GX01, GX08, GX09, GX014, GX019, GX091, GX123, GX173, GX174, GX195, GX260, GX443, GX452, GX510, GX520, GXBS, GXSL, GXC, G, GXLA, GXHX, GXGL, GXNniu, GXWXp, their homology of nucleotide was from 97.6 % - 100 %. Group Ⅱ includes 10 isolates GX074, GX219, GX304, GX441, GX442, GX496, GX0510, GXPX, GXBM, GXQZ, their homology of nucleofide was from 98.5 % - 100 %. Only GXN119 strain belongs to group Ⅲ. Group Ⅳ includes 8 isolates GXS06, GX508, GX509, GX120, GXB03, GXcat, GX201, GX102, their homology of nucleotide was from 99.1% - 99.6 %. According to the mutations of amino acid and compared with that of ERA, a vaccine strain for animal vaccination,the residue proline on position 58 become sorine in all 42 isolates. 34 isolates of group Ⅰ,Ⅱ,Ⅲ showed 6 common mutations H26Y,C40S,S61N,V95L,G106D and P135S. Group Ⅰ revealed 2 group - specific mutations T90N and E110D. Group Ⅱ revealed 1 group - specific mutation. These mutations of amino acid on nucleoprotein were coincidence with nucleotide homology,phylogenetic tree analysis. The rabies vires isolates from Guangxi were topographically distribution.
