摘 要:鉴定人胃癌细胞系BGC-823中的SP(side population)细胞,并进一步研究SP细胞对化疗药物5-氟尿嘧啶的耐受性。利用Hoechst33342对细胞染色,通过流式细胞仪检测及分选Hoechst低染的SP细胞;同样的方法检测5-氟尿嘧啶处理后的细胞中SP细胞比例的变化;MTT法检测分选后的SP与非SP细胞的抗药性差异。结果表明,人胃癌细胞系BGC-823中存在4%左右的SP细胞;细胞经过5-氟尿嘧啶处理后,SP细胞的比例显著升高;体外正常培养条件下SP细胞并不具备增殖优势,但分选后的SP细胞较非SP细胞显示出了更强的耐药存活能力。To identify the side population(SP)cells in human gastric cancer cell line BGC-823 and further study the chemotherapeutic drug resistance of SP cells to 5-fluorouracil. Cells are stained with Ho- echst 33342 and Flow Cytometry is used to detect and sort SP cells. After treatment with 5-fluorouracil, changes in the proportion of SP cells are detected using the same method. The differences in drug resistance between SP and non-SP cells are tested by MTT assay. Results. BGC-823 cell line contains about 4% SP cells. The proportion of SP cells increases significantly after 5-fluorouracil treatment. The sorted SP cells don't have the proliferation advantage in vitro, while they demonstrate the tougher viability than non- SP ones when suffering with drug treatment.
