摘 要:目的观察中药烧伤膏在烫伤创面愈合过程中转化生长因子β1(transforming growth factor-β1,TGF-β1)及微血管生成的影响,探讨其可能的作用途径。方法取28只SD大白鼠,制作深Ⅱ度烫伤创面模型,随机分为中药烧伤膏组和空白对照组,各组14只,记录创面愈合时间;分别在伤后第13、、5、7、10、14、20天取材,采用免疫组织化学SP法检测TGF-β1的表达,并计数阳性细胞数。以CD34一抗免疫组织化学标记血管内皮细胞,记数微血管密度。流式细胞术检测创面表皮细胞周期分布。结果中药烧伤膏组烫伤愈合时间明显早于对照组(P<0.01)。TGF-β1表达主要在炎症细胞和毛囊上皮细胞,伤后第3、第5天,中药烧伤膏组TGF-β1的阳性表达率明显高于对照组(P<0.01);中药烧伤膏组TGF-β1表达的高峰早于且强于对照组。第5天微血管密度记数中药烧伤膏组显著高于对照组(P<0.05)。细胞周期分析显示,创面经中药烧伤膏处理后,较对照组比较,出现细胞增殖活跃的时间明显提前。结论中药烧伤膏能促进烫伤创面的愈合,其作用机制可能与早期提高组织中TGF-β1的表达、促进新生血管的形成有关。Objective To observe the effect of Chinese materia mediea Shao Shang Ointment on transforming growth factor-β1, micrangium, and explore its possible effect pathway. Methods Twenty-eight Sprague-Dawley(SD) rats were used to make deep second-degree burn models and all these wounds were randomly divided into Chinese materia medica Shao Shang Ointment group and blank control group. Wound healing time was recorded; wound bed tissues were got on the 1st, 3rd, 5th, 7th, 10th, 14th and 20th days respectively after burns. Experssion of TGF-β1 was detected using immunohistochemistry method(SP), and counted the positive cells. Vascular endothelial cells were labeled with CD34- and-immunohistochemistry, and counted the density of micrangium. The distribution of epidermic cell cycle were detected by flow cytometry. Results Healing time in the Chinese materia medica Shao Shang ointment group was significantly earlier than that in the control group (10.0±1.2d, 15.0±2.2d,P〈0.01). The expression of TGF-β1 was at inflammatory ceils and epithelial cells of hair follicle. At the 3rd, 5th days after injury, the positive expression rate of TGF-β1 was signifincantly higher than that inthe control group(23.8±2.9)%,(10.1±2.8) %,(43.8±4.9) %,(23.8±2.9) %, P〈0.01. The peak of expression of TGF-β1 appeared earlier and stronger in the Chinese materia medica Shao Shang Ointment group than that in the control group. The counting of micrangium density at the 5th day was markedly higher in the Chinese materia medica Shao Shang Ointment group than that in the control group(44.8±4.1 piece, 33.5±5.9 piece, P〈0.05). FCM revealed that, compared with the control group, the athletic time of cell proliferation was more advanced in Chinese materia medica Shao Shang Ointment group.
