摘 要:环媒恒温核酸扩增(LAMP)反应,是分子生物学领域中的一个新概念,它可以在恒温(60—65℃)条件下,30—60m in内将只有几个拷贝的靶核酸扩增到109水平.该方法的一大特色是,可以通过反应副产物-白色焦磷酸镁沉淀的产生与否判断靶基因的存在.因此,对于病原微生物的鉴定具有高效率、高特异性、高敏感性等特点.本文重点论述了目前LAMP方法的原理以及在食品安全检测、流行性病毒检测、临床诊断以及水产养殖等领域的应用及研究情况.表明,该方法已具有成功应用的现实性,为快速基因检测提供了一种新的技术途径;比PCR方法更具推广性,可望作为常规检测工具.Loop-nlediated isothermal amplification of DNA (LAMP) is a novel concept in molecular biology. It can amplify a few copies of target nucleic acid to a magnitude of 109 under isothermal conditions in the range of 60-65 ℃ in 30-60min. The favorable peculiarity of this method is that it can demonstrate the absence of the target gene by the production of white precipitate of magnesium pyrophosphate. So the LAMP assay possesses the advantages of high efficiency, high specificity and high sensitivity. The main purpose of this paper was to summarize the application and research progression of LAMP in food safety inspection, detection of epidemic viruses, clinical diagnosis and aquaculture et al. The results indicated that LAMP had the realities of successful amplification and had more applicability than PCR. It provided a new technique route for rapid genetic diagnosis of pathogenic microorganism and could potentially be used as a conventional detection tool.
