摘 要:目的观察磺脲类药物格列美脲对2型糖尿病GK大鼠心肌组织ATP敏感性钾通道(KATP)表达的影响。方法自发性糖尿病GK大鼠48只随机分入糖尿病对照组、糖尿病胰岛素治疗组、糖尿病格列本脲组、糖尿病格列美脲组,另设正常对照组(n=12)。放射配体结合法检测SUR2蛋白含量,半定量RT-PCR法检测KATP通道亚单位SUR2和K ir6.2 mRNA表达。结果干预12周后糖尿病组、糖尿病胰岛素治疗组和正常对照组SUR2受体密度、SUR2 mRNA及K ir6.2 mRNA表达水平无显著差异;格列美脲治疗组SUR2受体密度较正常对照组和糖尿病组无明显改变;SUR2 mRNA及K ir6.2 mRNA表达水平无明显改变。第24周,糖尿病大鼠SUR2 mRNA表达水平较12周下降,也低于同期正常对照大鼠,但尚无统计学意义;格列美脲治疗组SUR2受体密度无明显改变;KATPmRNA表达水平变化不大。结论自发性糖尿病GK大鼠的糖尿病状态不影响心肌KATP通道表达水平;治疗剂量的格列美脲对糖尿病大鼠心肌组织KATP通道表达无明显影响。Objective To observe the effects of long-term treatment with Glimepiride on ATP-sensitive potassium channels (KATP )expression in myocardium of spontaneously diabetic Goto-Kakizaki (GK)rat, Methods Forty-eight spontaneously GK rats were divided into four groups : diabetes model group, positive control group( treated with insulin), Glibenclamide group and Glimepiride group. And a normal control group was also set up. The expression of SUR2 in myocardium of GK rat was investigated by radioligand binding assay, KATP mRNA expression in myocardial cells of rats was detected through reverse transcriptase-polymerase chain reaction(RT-PCR). Results had no differences between diabetes ( P 〉 0.05 ). Compared with control The SUR2 receptor density and mRNA expression of SUR2 and Kir 6.2 group, insulin treated group and control group after 12 weeks intervention group and diabetes group SUR2 receptor density and mRNA expression of SUR2 and Kir 6.2 had no significant differences in Glimepiride group. The expression of SUR2 in diabetes after 24 hours intervention were lower than that after 12 weeks intervention, and also lower than that in normal rats at the same time,but no differences were found. Conclusion With all Sulfonylureas-treated diabetic and non-treated diabetic rats it showed that there was no change in mRNA levels of KATP in heart ( P 〉 0.05 ). Conclusion The diabetes itself does not affect the ATP-sensitive potassium channels (KATP )expression in myocardial tissues. Glimepiride with treatment dosage has no effect on channels expression of KATP.
