摘  要:目的探讨泼尼松龙对实验性变态反应性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)中枢神经轴突损伤的影响以及与炎症反应及髓鞘脱失的关系。方法用MOG35-55蛋白多肽免疫C57BL/6J小鼠复制多发性硬化(multiple sclerosis,MS)的动物模型EAE,采用轴突的Bielschowsky染色结合常规形态学染色和神经系统髓鞘染色比较治疗组与对照组的差别。结果泼尼松龙能促进EAE临床症状的缓解;减轻中枢神经轴突损伤,同时能清除病灶中浸润的炎症细胞及降低神经髓鞘的脱失。但是当停止用药后,该疾病又复发,轴突的损伤与未治疗对照组相同。结论泼尼松龙能诱导实验性自身免疫性脑脊髓炎的缓解,对轴突损伤有短暂的保护作用。Objective To explore the effects of prednisolone on the protection of axonal damage in experimental autoimmune encephalomyelitis ( EAE), and the relationship of inflammatory responds with demyelination in the central nervous system. Methods Animal model of EAE, also as multiple sclerosis (MS) was established by immunizing C57BL/6J mice with myelin oligodendrocyte glycoprotein peptides35-55 ( MOG35-55 ) , and their clinical signs were scored. Totally 30 immunized C57BL/6J mice were divided into 3 groups, i. e. , untreated group, prednisolone continuously-treated group and prednisolone withdrawal group. Prednisolone at 60 μg/ml was added into the drinking water for the mice of the later 2 group at the onset of EAE ( 12 - 14 d after immunization). The drinking water was continued for the second group till the end of observation (55 d after immunization), but withdrawn for the mice of the third group at 3 d after the disappearance of symptoms. Axonal damage, inflammation and demyelination were observed by conventional histological method, Bielschowsky staining, and Luxol Fast Blue staining. Results Our data indicated that prednisolone could remit the clinical signs of EAE, attenuate axonal damage, eliminate the infiltrate inflammatory cells, and decrease demyelination in central nervous system. In prednisolone withdrawal group, the disease relapsed in 5 to 17 d after the drug stop, and the status of axonal damage was same as that of untreated group. Conclusion Prednisolone as a immune regulator can alleviate the signs of EAE and exert transitory protective effect on axonal damage.
