摘 要:目的为了探讨雷公藤治疗足细胞病变的机制,利用嘌呤霉素诱导足细胞凋亡的体外损伤模型,观察雷公藤甲素对足细胞损伤的保护作用及其机制。方法用CCK法观察雷公藤甲素对足细胞增殖活性的影响;应用TUNEL染色法定性观察雷公藤甲素对嘌呤霉素诱导的足细胞凋亡的保护作用;采用PI染色后再经流式细胞仪来定量分析雷公藤甲素对足细胞凋亡的抑制效应。结果雷公藤甲素质量浓度小于1ng/mL时对足细胞生存率没有明显影响;100μg/mL嘌呤霉素诱导足细胞凋亡,但是此凋亡效应可以被1ng/mL雷公藤甲素抑制;而且这种抑制作用具有时间依赖性。结论雷公藤甲素通过抑制细胞凋亡来保护足细胞。雷公藤甲素对足细胞的保护作用可能是临床上雷公藤有效治疗足细胞病变的机制之一。Objective To explore the therapeutic effects of triptolide on pm'omycin aminonucleoside (PAN)-induced podocyte injury and its mechanism. Methods Podocytes were treated with various concentrations of triptolide (0, 0.5,1, 2, 4, 8 ng/mL). The cell viability was measured by Cell Counting Kit-8 assay; apoptotic podocyte cells were identified by terminal deoxynucleotidyl transferase-mediated deoxy- UTP nick end labeling (TUNEL) assay; the apoptotic rate was detected by flow cytometry after staining podocyte cells with PI. Results Triptolide ( 〈 1 ng/mL) did not decrease the cell viability significantly. After treatment with 100 μg/mL PAN for 24 h, the apoptotic rate of podocytes increased dramatically compared with control [ (20.11± 0.41 ) % vs ( 0.29 ± 0.18 ) % ]. Tfiptolide ( 1 ng/mL) could remarkably inhibited the PAN-induced apoptosis ( 10.46 ±0.4) % in a time-dependent manner. Conclusion Triptolide has obviously protective effects on podocyte, which may be related to the inhibition of cell apoptosis. The results suggest that the anti-proteinuria effects of triptolide in clinic might due to its direct protective effects on podocyte.
