摘 要:紫膜蛋白菌紫质(bacteriorhodopsin,BR)处于不同的膜脂环境中,通过吸收光谱、荧光光谱和闪光动力学光谱的测定,比较了3种不同膜脂环境对菌紫质分子结构和功能的影响。实验结果表明:天然膜脂是BR最稳定的膜脂环境,可以形成以三聚体为单位的二维六角形晶格结构。二豆蔻酰脂酰胆碱(dimyristoyl phosphatidylcholine,DMPC)也是BR分子的稳定脂环境,但以单体形式存在于膜脂环境中,功能受到一定影响。经TritonX-100增溶处理的紫膜膜脂环境中BR为单体,不稳定,容易发生结构和功能的变化。Bacteriorhodopsin (BR) was lacated in three different lipid environments, the effect of lipid environments on the structure and function of bacteriorhodopsin was studied by absorbance spectra, fluorescence spectra and flash dynamics spectra. The results showed that native membrane lipid was the most stable membrane lipid environments, bacteriorhodopsin was arranged in a two-dimentional hexagonal crystal lattice by trimer. Dimyristoyl phosphatidylcholine (DMPC) also was stable membrane lipid environments, bacteriorhodopsin was appeared to be membrane in monomeric state, the function of bacteriorhodopsin of monomeric state was effected by DMPC membrane lipid environments. TritonX-100 dissolved BR monomer was not stable, the structure and function of bacteriorhodopsin was changed in this lipid environments.
