摘  要:目的:观察萘哌地尔(NAF)衍生物YMO1对正常大鼠血压的影响,对其降压效果进行评价.方法:采用SD大鼠,分别静脉给药及十二指肠给药,观察YMO1对大鼠血压和心率的影响.结果:YMO10.5,1,2mg/kg静注均可使SD大鼠血压降低,且降压效应呈剂量依赖性,十二指肠给予YMO11,5,10mg/kg,连续观察2.5h,血压均有不同程度的降低,心率却无明显变化.结论:①NAF衍生物YMO1有明显的降压作用,YMO1具有与其母核NAF类似的心血管药理作用;②YMO1起效平缓稳定,口服有效.AIM: To observe the effects of naftopidil derivative YMO1 on the blood pressure and heart rate in rats. METHODS: The effects of YMO1 on blood pressure and heart rate in SD rats were measured after YM01 was given intravenously or intragastrically. RESULTS: Intravenous administration of YMO1 (0.5, 1, 2 mg/kg) decreased the blood pressure in rats at a dose-dependent manner. Intragastrical administration of YMO1 ( 1, 5 and 10 mg/kg ) decreased the blood pressure in rats during 2 h ,however, the heart rate was not influenced significantly by YMO1. CONCLUSION : ( 1 ) The derivative YMO1 of naftopidil can obviously decrease blood pressure, and possesses similar cardiovascular pharmacological action with naftopidil. (2) The efficacy of YMO1 is gentle and steady after they have been absorbed by oral.
