摘 要:采用肉眼观察法测定不同5-羟色胺(5-HT)受体拮抗剂对日本血吸虫运动性的影响。结果表明:(1)10-3mol/L和10-4mol/L的5-HT作用虫体30min后,虫体运动显著增强,而10-5mol/L时,虫体运动稍有增强。10-3mol/L色胺(tryptamine)也可显著增强虫体运动,但10-4mol/L、10-5mol/L时,作用则不显著。(2)用10-3mol/L赛庚啶(cyproheptadine)、曲拉唑酮(trazodone)、米安舍林(mianserin)、螺环哌啶酮(spiroperidol)、氟哌丁苯(haloperidol)作用虫体1h后,除haloperidol外,其余均可对虫体产生不同程度的拮抗作用。(3)先用10-4mol/L5-HT培养虫体30min,当虫体运动增强后,再用上述拮抗剂作用虫体1h,发现cyproheptadine、mianserin、trazodone对虫体有拮抗作用,spiroperidol略有拮抗,而haloperidol有一定协同兴奋作用,这些拮抗剂不仅对内源性5-HT有作用,而且对外源性5-HT均有拮抗性。初步判定,日本血吸虫体内存在5-HT受体。Motor activity was determined with drugs at different concentration under a microscope. The results showed that S. japonicums were obviously stimulated by 5hydroxytryptamine (10 -3 , 10 -4 mol/L) and tryptamine (10 -3 mol/L). Several vertebrate 5hydroxytryptamine antagonists, cyproheptadine, mianserin and trazodone, at concentration around 10 -3 mol/L not only blocked the effects of endogenous 5HT but also reduced exogenous 5HTinduced increases in the motor activity of S. japonicum . Spiroperidol, the other 5HT antagonist, had no influence on the motility. To our interest, haloperidol had coordinated excitation all along. The above effects preliminarily suggested the presence of 5HT receptor in S. japonicum and it′s difference from mammalian′s.
