Effect of Ebselen on NO/NOS System and Free Radical Metabolism Induced by High-Intensity and Endurance-Training in the Rat

摘  要:目的:探讨ebselen对运动训练大鼠肾脏丙二醛、总抗氧化能力、超氧化物歧化酶、谷胱甘肽过氧化物酶以及一氧化氮和一氧化氮合酶表达的影响。方法:SD雄性健康大鼠30只,随机分为3组:安静对照组、运动对照组、运动加药组各10只;后两组进行为期8周递增负荷跑台训练,建立大强度耐力训练模型测定肾脏上述相应指标。结果:用ebselen干预后,相比运动对照组,运动服药组各项指标均有明显改善。丙二醛含量显著降低(P<0.05),总抗氧化能力显著升高(P<0.05),谷胱苷肽过氧化物酶活性显著升高(P<0.05),超氧化物歧化酶活性显著升高(P<0.05),一氧化氮合酶和诱导性一氧化氮合酶表达有所下降(P<0.05),NO生成减少(P<0.05)。结论:ebselen对大强度耐力训练中肾脏产生的自由基有良好的清除作用,可以有效对抗运动造成的肾脏缺血再灌注损伤。Objective: To study the effect of ebselen on the expression of MDA, SOD and GSH-Px, total antioxidant capacity, and the expression of NO and NOS system in the trained-rat's kidney. Methods: Thirty male healthy SD rats were randomized into 3 groups, the control group (A), exercise group (B) and exercise added ebselen group (C) with 10 rats in each group. Group B and C was arranged eight weeks' training and establish the high-intensity and endurance-trained model for 8 weeks. The contents of MDA, SOD, GSH-Px, TAOC, NOS, iNOS and NO were tested in rat's kidney. Results: After being interfered with ebselen, there were significantly improvement in the various index compared with exercise group, the contents of MDA and NO decreased significantly(P〈0.05), the concentration of SOD and GSH-Px increased markedly(P〈0.05), the activity of iNOS and NOS decreased notably(P〈0.05), the production of NO reduced obviously (P〈0.05). Conclusion: This result indicated that ebselen had a good effect in removing the free radical in rat's kidney during high-intensity and endurance-trained and ebselen has significant protective effect on kidney of ischemical reperfusion injury.
