特发性血小板减少性紫癜患者T细胞周期和促凋亡蛋白Caspase-3表达的变化

摘　　要：目的:探讨特发性血小板减少性紫癜(ITP)患者外周血T细胞的细胞周期及其促凋亡蛋白Caspase-3的表达在发病机制中的作用。方法:用流式细胞仪检测38例ITP患者和20例正常人的T细胞的细胞周期的分布比例及30例ITP患者和20例正常人的T细胞凋亡蛋白Caspase-3的表达水平。结果:ITP患者外周血S期和S+G2/M期的T淋巴细胞比例显著高于对照组(P<0.05);Go/G1的T淋巴细胞比例显著低于对照组(P<0.05);T淋巴细胞Caspase-3的表达率明显低于对照组(P<0.05)。结论:在ITP患者中存在T淋巴细胞过度增殖,细胞以S,G2期为主,同时T淋巴细胞存在凋亡减少,这可能与ITP的发病有关。Objective： To explore the molecular regulatory mechanism of the cell discribution and the expression of caspase 3 in peripheral blood T lymphocytes of idopathic thrombocytopenic purpura （ITP） patients. Methods： Distribution of cell cycle of T lymphocytes from 38 patients with ITP and 20 normal subjects and expression of caspase-3 from 30 patients with ITP and 20 normal subjects was analyzed by flow cytometry. Results： The percentage of T lymphocytes in the S and S＋ G2/M phase was significantly higher, and the percentage in the G0/G1 phase was lower than that in the control group, respectively （all P〈0.05）. The expression of caspase-3 in T lymphocyte of the ITP group was significantly lower than that of control group （P〈0.05）. Conclusion： T lymphocytes of the ITP patients in proliferation stage are increased, mostly in S and G2/M phase, and the apoptosis T lymphocyte number is decreased. The abnormities of proliferation and apoptosis probably participate in the pathogenesis of ITP.
