不同生长环境中博落回生物碱含量及提取工艺研究

摘　　要：目的:确定博落回在不同生长环境中生物碱的含量,为进一步开发利用博落回植物资源,提供科学依据。方法:采用正交试验,以浸膏得率和生物碱含量为考察指标,对博落回生物碱的提取条件进行优选。然后,在该条件下对同一时期采集的不同生长环境中的博落回进行生物碱提取及其含量的测定。结果:各考察因素对博落回生物碱提取工艺的影响程度为乙醇溶液的pH值>提取时间>溶剂量倍>提取次数。pH值为2的85%醇溶液中浸18h,固液比20倍量、回流提取3次、每次40min为最佳工艺条件。博落回野生植株生物碱含量大于栽培的。结论:博落回生物碱提取的最佳条件为A_1B_2C_3D_1;博落回栽培植株与野生的生物碱含量是有差异的。Objective： To ascertain the alkaloid content of Macleaya cordata in different growing environments to provide scientific basis for further expoitation and utilization on its resourses. Methods： The optimum extraction of alkaloid was investigated by orthogonal design as the alkaloi and total mount of dry ointment content index, alkaloids of Macleaya cordata adopted same times in different growing environments were extracted and determined by this optimum condition. Results： extraction process was affected each factor in the proportion as follows - pH value of ethanol concentration 〉 extraction duration 〉 solvent volume 〉 extraction times. The best condition is to soak 18 hours into pH value 2 of 85% ethanol with 20-fold solid-liquid ratio, reflux extraction for 3 times and 40 minutes for each time. The alkaloid content from the wild plant of Macleaya cordata is more than the cultivated one. Conclusion： The best condition for extraction of alkaloid is A1 B2 C3 D1 ; there is some difference in the content of alkaloid of the cultivated plant and the wild one of Macleaya cordata.
