摘 要:目的从旋毛虫感染的大鼠和兔血清中纯化抗旋毛虫IgG抗体。方法分别用辛酸-硫酸铵法(CA-AS)、硫酸铵沉淀法(AS)纯化旋毛虫感染的大鼠和兔血清中抗旋毛虫IgG抗体。结果IgG抗体纯化率CA-AS法为80.05%~83.22%,AS法为67.00%~71.08%;得率CA-AS法为7.85~8.79mg/ml,AS法为12.04~12.12mg/ml;回收率CA-AS法为52.55%~55.03%,AS法为61.36%~64.87%。CA-AS法得率和回收率均低于AS法,但纯化率高于AS法。ELISA法测定CA-AS法和AS法纯化的抗体效价不变。结论CA-AS法是一种简便?快速?有效的纯化血清IgG抗体的方法,可用于大鼠和兔血清抗旋毛虫IgG抗体的纯化。Objective To purify anti-Trichinella spiral IgG antibodies in sera from rat and rabbit infected with T.spiral.Methods To purify anti-T.spiral IgG antibodies in sera from rat and rabbit infected with T.spiral by caprylic acid-ammonium sulfate precipitation(CA-AS)and ammonium sulfate precipitation(AS),respectively.Results Purity of IgG ranged from 80.05% to 83.22% by CA-AS,while ranged from 67.00% to 71.08% by AS;Yield of IgG ranged from 7.85% to 8.79% by CA-AS,while ranged from 12.04% to 12.12% by AS;Recovery of IgG ranged from 52.55% to 55.03% by CA-AS,while ranged from 61.36% to 64.87% by AS.Yield and recovery of IgG antibody all were lower by CA-AS than those by AS,while purity was higher by CA-AS than that by AS.There were not difference in the titer of serum and IgG by ELISA.Conclusion CA-AS was one simple,rapid,effective method for purification of IgG antibody in serum,and could be used for purification of anti-T.spiral IgG antibody in sera from rat and rabbit infected with T.spiral.
