健胃愈疡颗粒对大鼠胃黏膜癌变DNA含量、TNF-α基因表达及细胞凋亡的影响

摘　　要：目的:研究健胃愈疡颗粒加维甲酸对大鼠胃黏膜癌变DNA含量、TNF-Α基因mRNA表达及细胞凋亡的影响。方法:采用MNNG、酒精及10%NaCl混合液建立大鼠胃癌模型,设立正常空白组、模型组维甲酸治疗组和健胃愈疡颗粒加维甲酸治疗组,分别采用Feulgen染色、免疫组化,Western blotting及TUNEL法检测大鼠胃黏膜DNA含量、TNF-α基因表达及细胞凋亡。结果:健胃愈疡颗粒加维甲酸治疗组癌变发生率低于模型组和维甲酸治疗组。TNF-α基因表达与细胞凋亡受抑制和胃黏膜癌变的发生,呈递增趋势,并且两者具有一定的相关性。结论:健胃愈疡颗粒加维甲酸抗癌,下调TNF-α基因表达,促进细胞凋亡是其重要机制之一。AIM ： To study the effects of Jianwei Yuyang granule in combination of transretinoic acid on DNA, TNF-α exepression and apoptosis of gastric cancer in rats. METHODS： We studied DNA, expression of TNF-α gene and apoptosis using feulgen, in situ hybridization, TUNEL to set up rat＇s gastric cancer model experiment rats were assigned to 4 groups ： 1. normal control, 2. model control, 3. transretinoic acid reatment Groups, 4. Jianwei Yuyang Granule combined with transretinoic acid tratnment groups in gastric cancer model induced by MNNG, alcohol and 10 % sodium chloride. RESULTS： The incidence of gastric cancer in Jianwei Yuyang Granule in combination with transretinoic acid treatment groups was lower than that of model ones and transretinoic acid groups. With the development of gastric cancer , the expression of TNF-α gene and supression of apoptosis increased. A corrolation was present between both. CONCLUSION： Jianwei Yuyang Granule in combination with transretinoic acid can both downregulate the expression of TNF-α gene and promote cell apoptosis in experiment rats.
