摘 要:应用双向凝胶电泳技术(two-dimensional gel electrophshiya/oresis,2-DE),比较橡胶树死皮株与健康株胶乳黄色体蛋白质组表达的差异。采用TCA/丙酮沉淀法提取橡胶树死皮株与健康株黄色体蛋白质,并采用固相pH梯度(immobilized pH gradient,IPG)双向凝胶电泳分离两种材料蛋白质,凝胶经银染显色后,用PDQuest图像分析软件进行比较分析、识别差异表达的蛋白质。成功获得了橡胶树死皮株与健康株胶乳黄色体的双向凝胶电泳图谱。鉴定出24个蛋白差异点,其中17个上调表达,7个下调表达。并应用质谱技术鉴定了其中部分表达差异的蛋白质点,对渗调蛋白进行了功能分析,其在死皮株中表现下调的情形可能与死皮病的发生有一定的关系。Differential proteomic analysis was conducted in lutoid proteins from latex of healthy and Tapping panel dryness (TPD) trees by two-dimension gel electrophoresis(2-DE).The lutoid proteins from latex of healthy and TPD trees were isolated respectively by TCA/cold acetone precipitation. And these were prepared by means of immobilized pH gradient (IPG) based on 2-DE. After silver staining, the differential expression proteins of two kinds were analyzed using image analysis software (PDQuest). Using proper method stated above, and satisfactory 2-DE maps of lutoid proteins from latex of healthy and TPD trees were obtained and the preliminary analysis results were reported. There were 24 notable spots were detected, including 17 up-regulated proteins and 7 down-regulated proteins. Of them some different-expressed protein spots were detected by mass spectrometry. There were some relationship between osmotin protein with TPD broom according to the functions of down-regulated proteins in TPD trees.
