健胃愈疡颗粒剂对大鼠胃溃疡模型溃疡愈合及胃组织IL-1β蛋白表达的影响

摘　　要：目的:探讨健胃愈疡颗粒对大鼠胃溃疡模型溃疡愈合及胃组织IL-1β蛋白表达的影响。方法:用Okabe改良法复制SD大鼠胃溃疡模型,观察溃疡指数,采用免疫组织化学法观察胃粘膜组织形态及IL-1β蛋白表达的变化。结果:①健胃愈疡组的溃疡指数明显低于模型组和法莫替丁组;②HE染色法显示健胃愈疡组较模型组及法莫替丁组黏膜厚度增加,溃疡床及溃疡周围炎性细胞浸润明显减少,纤维排列较为整齐。③IL-1β在正常胃组织呈阴性或偶有阳性信号表达,造模型7 d后表达显著增加,健胃愈疡组和法莫替丁组低于模型组(P<0.01);14 d后各组的阳性信号表达均降至接近正常。结论:健胃愈疡颗粒能保护大鼠胃粘膜组织,降低胃溃疡大鼠模型溃疡指数,改善溃疡愈合质量,减少胃组织IL-1β蛋白的表达,促进溃疡愈合。Objective：To investigate the effects of J ianweiyuyang granules（JWYYG）on ulcer healing and the expression of IL-1βin gastric tissue of gastric ulcer（GU）model in rats.Methods：100 SD rats were randomly divided into five groups：normal group （n=20）,sham surgery group（n=20）,famotidine group（n=20）,JWYYG group（n=20）and model group（n=20）prepared by modified Okabe method.The expression of IL-1βwas determined by immunohistochemistry.Results：The ulcer index of JWYY group was significantly lower than that of model group and famotidine group（P〈0.01）.The expression of IL-1βprotein in GU model group was higher than that in normal group and famotidine group,while the positive expression of IL-1βprotein in each group nearly dropped to normal level.Conclusion：Jianweiyuyang granules can protect gastric tissue and accelerate ulcer healing by decreasing ulcer index of gastric ulcer rats model and expression of IL-1βprotein in the gastric tissue.
