摘 要:目的:本文在体外条件下观察了罗氏海盘车体腔细胞的形态特征。方法:采用镜下观察和化学发光法并在液体闪烁仪(LS-5801)检测。结果:观察发现罗氏海盘车的体腔细胞可分成:大颗粒细胞(桑椹细胞)、小颗粒细胞、大透明细胞、小透明细胞、柳叶形细胞、兼性大细胞和杆状颤动体;体腔细胞的浓度范围是:5.62~10.00×10~6个/mL。吞噬观察中发现小颗粒细胞伪足明显、吞噬力强,在不同温度下吞噬率和吞噬指数不同,14℃下最高,分别为:62.6%和2.06.体腔细胞在吞噬过程中也会产生呼吸爆发现象,产生活性氧(O_2^-)、(H_2O_2)、(OH.)、(~1O_2)等。在适宜条件下,体腔细胞吞噬酵母的化学发光规律随时间移动为一条有单峰值的曲线;在不同环境条件下,pH=6.81时,化学发光最明显;在外界诱导物刺激下,酵母悬液浓度为1.00×10~7cell/mL时,化学发光最明显。结论:罗氏海盘车体腔细胞发挥了重要的免疫功能。Objective:To observe the characteristic shape of the coelomocyte of Asterias rollestoni Bell.Methods:microscope and the method of chemiluminescence measuring were used.Results:The coelomocyte could be divided into 7 types:large grained cell,small granular cell,large hyaline leucocyte,small hyaline leucocyte,lanceolate cell,facultative cell and bacillary flutter.The concentration range of the coelomocyte was 5.62~10.00×10^6/ml.In the observation of phagocytize,pseudopod were obviously found and the phagocytic power was strong in the small granular cell.Different temperature has different phagocytic rate and phagocytic index,When temperature reached 14℃,it would be reached peak value,62.6 % and 2.06 respectively.During the course of phagocytizing of coelomocyte, there would be breathing breakout with(O2),(H2O2),(OH.)and)(1^O2)appearance.The lightening rule of the coelomocyte phagocytize yeast were curve with unimodal titre by time retarting in proper condition.In different conditions,such as when pH reached 6.81,the chemiluminescence was most obviously,also when the yeast suspension concentration was 1.00×10^7cell/ml.Conclusion:The coelomocyte of Asterias rollestoni Bell played important immune function
