不同激素添加成分和受精液对山羊卵巢卵母细胞体外成熟与受精的影响  

Effects of Different Hormone Supplement Components and Fertilization Medium on in vitro Maturation and Fertilzation in Goat Ovary Oocyte

摘　　要：就成熟培养液中添加成分和不同受精液对山羊卵母细胞体外成熟与体外受精的影响进行了研究。结果表明,成熟培养液中添加HCG组的卵母细胞成熟率(85.7%)极显著高于PMSG组(50%)和FSH+LH组(57.1%)(P<0.01),受精率比较则HCG组(62.2%)极显著高于PMSG组(34.7%)(P<0.01),显著的高于FSH+LH组(45.2%)(P<0.05);添加了17β-雌二醇的成熟培养液对山羊卵母细胞的成熟率和受精率没有明显影响(P>0.05),而添加5%山羊卵泡液的成熟培养液则使卵母细胞的成熟率和受精率明显下降,卵裂率(7,7%)却显著高于对照组(0.0%)和17β-雌二醇组(0.0%)(P<0.05);分别以TALP液、BO液、M-199液为基础液组成的三种受精液(Ⅰ液、Ⅱ液、Ⅲ液)中Ⅱ液的受精率(62.2%)显著高于Ⅲ液(44.7%)(P<0.05),而且Ⅱ液的卵裂率(21.6%)显著高于Ⅰ液(9.1%)(P<0.05),极显著高于Ⅲ液(0.0%)(P<0.01)。The effects of supplement components in the maturation culture medium of oocytes and different fertilization medium on goat oocyte maturation and fertilization in vitro were studied. The results showed that the oocytes maturation rate in the treatment inwhich HCG（85.7% ） was supplimented into the maturation culture medium was extremly significantly higher than those in the PMSG group（50% ） and FSH ＋LH group（57.1% ） （P 〈 0.01 ）, the fertilization rate of the HCG group （62.2%） was significantly higher than those in the PMSG group（ 34.7% ） （P 〈0.01 ） and FSH ＋ LH group （45.2%） （P 〈0.05） ; 17β - estradiol supplement into the maturation culture medium had no obviously influence on the maturation and fertilization rate in goat oocytes, but when 5% GFF was supplemented into the maturation culture medium, the rates of oocyte maturation and fertilization were obviously descended, on the contrary, it＇ s cleavage rate（7,7% ） was significantly higher than those of the control group （0.0%） and the 17β - estradiol group （0.0%） （ P 〈 0.05 ）, compared with the three fertilization mediumso（ Ⅰ, Ⅱ and Ⅲ ）in which TALP, BO and M -199 medium were used as the basic medium,the fertilization rate on mediusoⅡ（62.2% ） was significantly higher than that in medium Ⅲ（44.7% ） （P 〈0.05） and it＇s cleavage rate（21.6% ） was significantly higher than that on medium Ⅰ（9. 1% ） （P 〈0.05）and medium Ⅲ （0.0% ） （P〈0. 01）.
