摘  要:目的采用溶剂蒸发法制备聚乳酸(PLLA)微球。方法通过正交实验设计优化PLLA微球制备工艺考察了搅拌速度、PVA浓度、PLLA浓度、N2流速、针头直径对评价指标(即微球形态、粒径大小、粒径分布、分散性)的影响,确定制备不同粒径微球的最佳工艺条件。结果采扫描电子显微镜观察微球的外观形态,微球平均粒径为20μm且粒径分布集中。5因素对评价指标影响的主次顺序分别为:搅拌速度、PVA浓度、N2流速、PLLA浓度、针头直径。结论该经优化制备的PLLA微球分散性和成球性好,为下一步载药微球的制备提供了基础。Aim To prepare PLLA microspheres by means of the solvent evaporation. Methods The influences of different factors on microspheres quality were evaluated by orthogonal-designing method. Five factors were considered in the experiment, which were stirring rate, PVA concentration, PLLA concentration, N2 velocity and needle gauge, respectively. Four indexs were evaluated by microspheres quality, which were particle size, appearance, distribution of particle size and decentralization. Results The microspheres prepared by the optimum preparation method were smooth, regular in morphology, good decentralization, and the average particle size was 20um with narrow size distribution. Conclusion The important order of the factors that effect microspheres quality was stirring rates, PVA concentration, N2 velocity, PLLA concentration, needle gauge.
