摘  要:目的探讨电化学法对前列腺增生的影响及作用机制。方法将SD大鼠去势7d后皮下注射睾酮4mg/kg,于给药后20d时给予前列腺腔内介入电化学治疗,介入术后10、20、30d时分别处死大鼠,观察前列腺重量、前列腺尿道面及其邻近器官形态和组织结构损伤情况。通过免疫组织化学法检测增殖细胞核抗原(PCNA)和Bcl-2蛋白表达的改变。结果电化学治疗组大鼠前列腺重量与假手术组相比,差异有统计学意义。光镜证实电化学法可破坏前列腺上皮细胞,使细胞变性坏死。免疫组化法证实电化学治疗组PCNA指数与假手术组相比,差异有统计学意义(P<0.01)。而Bcl-2蛋白表达各组间差异无显著性(P>0.05)。结论电化学法可通过破坏前列腺上皮细胞抑制细胞增生,从而发挥治疗前列腺增生的作用,为电化学法临床应用提供了实验依据。Objective To study the effect of electrochemistry therapy (ECT) on animal with benign prostatic hyperplasia and its mechanism. Methods Castrated SD rats were administered exogenous testosterone by subcutaneous injection and then given electrochemistry therapy 20 days after that. The rats were decapitated at 10,20 and 30 days respectively after ECT, the weight of their prostate and injury condition of tissue structure and morphous of prostatic urethral surface and its neighbored organs were observed. Expressional changes of PCNA and Bcl-2 protein were analyzed by immunohistochemistry. Resets There is difference with statistical significance between group of electrochemistry theray and group of sham operation in the weight of rat prostate. Data from light microscope showed that electrochemistry therapy could destroy the prostatic epithelium, and resulted in cell degeneration and necrosis. Data from immunochemistry showed that there is statistic significanceBetween group of electrochemistry therapy and group of sham operation in PCNA index (P 〈0.01 ). There is no statistic difference about Bcl-2 expression (P 〉 0.05 ), Conclusion ECT can play its role in treating prostatic hyperplasia by destroying the prostatic epithelium and inhibiting cellular proliferation,which will provide experimental reference for clinical application of ECT.
