甘草酸二铵对慢性病毒性心肌炎小鼠心肌的保护作用

摘　　要：目的:探讨细胞间黏附分子-1(ICAM-1)、白细胞介素-2(interleukin-2,IL-2)及其受体(interleukin-2 receptor,IL-2R)在慢性病毒性心肌炎过程中的作用及甘草酸二铵(diammonni glycyrrhizinatis,DG)的影响。方法:建立慢性病毒性心肌炎小鼠模型,DG干预后,取心肌观察其组织病理变化;用逆转录-聚合酶链反应(RT-PCR)法和免疫组织化学方法检测ICAM-1及其蛋白表达水平,酶联免疫方法测定血清IL-2及可溶性白细胞介素-2受体(sIL-2R);酶化学法测定血清肌酸肌酶(creatine ki-nase,CK)活性。结果:慢性病毒性心肌炎小鼠心肌扭曲、细胞肿胀,ICAM-1表达水平、血清IL-2、sIL-2R水平及CK活性升高,DG可减轻心肌细胞的肿胀和扭曲,抑制慢性病毒性心肌炎ICAM-1表达、血清IL-2、sIL-2R水平及CK活性的升高。结论:ICAM-1、IL-2及其受体参与、影响慢性病毒性心肌炎的炎症过程,DG可通过抑制ICAM-1的表达、血清IL-2、sIL-2R的升高而产生心肌保护作用,在慢性病毒性心肌炎的防治中可能有一定的意义。OBJECTIVE To investigate the influence of expression of intercellular adhesion molecule-1 （ICAM-1）, interleukin-2 （IL-2） and soluble interleukin-2 receptor（sIL- 2R） on chronic stage of viral myoearditis, preliminarily probe the feasibility of drug intervention. METHODS The mice model with VMC was established. Myocardial histology was observed. Expression of ICAM-1 mRNA and protein was detected by using semi-quantitative RT-PCR and immunoeytoehemistry method. The IL-2 and sIL-2R levels in serum were examined by EIISA method and CK activity of serum were examined. These results were compared with those after diammonium glyeyrrhizinate （DG） intervention. RESULTS Myoeardium was twist and myocardial cell was swelling in mice with VMC. Myocardial levels of expression of ICAM-1 mRNA was increased. The ICAM-1 protein, IL-2 and sIL-2R levels in serum were also increased, but this condition in DG group was improved in some extent. CONCLUSION The findings indicate that IL-2, sIL- 2R and ICAM-1 is implicated in the development of chronic stage of viral myoearditis. DG has potential treatment value for chronic stage of viral myoearditis by partially blocking the expression of ICAM-1.
