摘 要:目的研究固相合成的Hymenistatin-1〔以下简称HS-1,序列为cyclo-(-Pro-Pro-Tyr-Val-Pro-Leu-Ile-Ile-)〕及其四种statine类似物的免疫活性。方法通过DNFB诱导小鼠迟发型超敏反应试验、体外淋巴细胞增殖试验和小鼠碳廓清试验,研究HS-1及其类似物对小鼠迟发型超敏反应、体外淋巴细胞增殖和巨噬细胞吞噬能力的影响。结果HS-1及其四种类似物对淋巴细胞增殖都表现出抑制作用,且修饰后的HS-1类似物在低浓度时表现出比阳性对照和天然HS-1更强的免疫抑制作用;A3 HS1低浓度组(50μg/ml)对淋巴细胞增殖的抑制作用最强,抑制率为28.4%。用statine修饰的HS-1类似物A2 HS1和A4 HS1能够抑制巨噬细胞的吞噬功能,但不能对小鼠迟发型超敏反应有很好的抑制作用。通过胸腺指数和肿胀度的比较可以得出,阳性对照在高浓度的时候对迟发型超敏反应有抑制作用,HS-1类似物的低浓度组比高浓度对小鼠迟发型超敏反应的抑制作用强。其中A1 HS1和A3 HS1的低浓度组已经表现出比天然HS-1组和阳性对照组强的免疫抑制活性。结论HS-1类似物A2 HS1和A4 HS1对巨噬细胞吞噬能力的抑制作用和对淋巴细胞增殖的抑制作用比阳性对照组和天然HS-1组强;HS-1类似物A1 HS1和A3 HS1对小鼠迟发型超敏反应的抑制作用和对淋巴细胞增殖的抑制作用比阳性对照组和天然HS-1组强。由此可见,通过对天然HS-1进行statine修饰,能增强其免疫抑制作用。Objective To study the Immunological activity of cyclopeptide Hymenistatin-1 and its analogues. Methods With the delayed type hypersensitivity reaction (DTH) induced by DNFB,LPT(lymphocyte proliferation test) and carbon clearance test to identify the effects of HS- 1 and its analogues on DTH reaction, lymphocyte proliferation and macrophage function of mice. Results HS-1 and its four analogues can inhibit the lymphocyte proliferation, and the modified HS-1 analogues have a stronger immunosuppressive effect titan the CsA group and native HS-1 group at low concentration; Low concentration group of A3HS1 has the highest inhibitory effect on lymphocyte proliferation, the inhibition ratio is 28.4% .Analogues A2HS1 and A4HS1 have inhibitory effects on phagocytosis of macrophage, but can not inhibit DTH reaction of mice. Contrast with the CsA group, all the analouges of HS-1 have a stronger inhibitory action on DTH at low concentration than that at high concentration. The low concentration groups of A1HS1 and A3HS1 showed more immunosuppressive effects than the native HS-1 and CsA groups. Conclusion A2HS1 and A4HS1 have a higher immunosuppressive effect on phagocytosis of macrophage and lymphocyte proliferation than native HS-1 and CsA. A1HS1 and A3HS1 can inhibit the DTH and lym.phocyte proliferation more effectively than native HS- 1 and CsA.So,the modification of HS-1 with statine can increase the immunosuppressive actions of native HS-1.
