八肋游仆虫核糖体蛋白L11的基因克隆与序列分析

摘　　要：以单细胞真核生物八肋游仆虫Euplotes octocarinatus为实验材料,采用PCR、RT-PCR方法克隆了核糖体蛋白L11基因(EoRPL11)。将该序列与GenBank中其他物种的RPL11基因序列进行同源性比对,采用DNAStar软件进行聚类分析。结果显示,成功克隆到游仆虫RPL11基因,大核中该基因全长709bp,开放阅读框(ORF)为531bp,编码176个氨基酸;cDNA序列与大核中ORF序列一致,表明该基因具有转录活性;所推导的氨基酸序列与嗜热四膜虫Tetrahymena thermophila的RPL11同源性最高,为66%。The gene EoRPL11 encoding the ribosomal protein L11 in Euplotes octocarinatus was amplified by PCR and RT- PCR. The deduced amino acid sequence of EoRPL11 was aligned with other RPL11s from different organisms. Phylogenetic relationship between EoRPL11 and some other RPL11 s was analyzed by DNAStar. The results showed that： the EoRPL11 gene in macronuclear DNA was 709 bp, containing an open reading frame （ORF） of 531 nucleotides which encoded a putative protein of 176 amino acids. Transcriptional activity of EoRPL11 was confirmed by RT-PCR. The deduced amino acid sequence shared 66% identity with RPL11 from Tetrahymena thermophila.
