摘 要:目的考察8种不同型号的大孔吸附树脂对多被银莲花总皂苷的吸附性能。方法采用静态吸附方法确定适合的树脂型号;采用动态吸附方法,以总皂苷的得率为指标,确定工艺条件。结果HPD400型大孔吸附树脂对多被银莲花总皂苷具有良好的纯化富集作用,其工艺条件为:树脂药材质量比2∶1,上样液质量浓度为每10 mL含1 g药材,以10 BV.h-1的流速上样,洗脱溶剂用体积分数为70%的乙醇。结论用HPD400型大孔吸附树脂富集多被银莲花总皂苷所得提取物纯度较高,总皂苷得率75%以上,此工艺条件可行。Objective To set up the preparation technology for purifying and enriching total saponin in rhizome of Anemone raddeana Regel by macroporous adsorption resin, and to investigate eight various types of macroporous adsorption resin. Methods The static adsorption and desorption were used in selecting the type of macroporous adsorption resin; with dynamic adsorption and desorption, the yield of total saponin were used as the evaluation criteria to investigate conditions of purification and enrichment. Results HPD 400 was the best type for purification and enrichment of the total saponin in rhizome of A. raddeana in the following technological condition: the ratio of the resin to crude drug was 2:1; the concentration of the raw material was 1 g crude drug in 10 mL extract; the flow rate of adsorption was 10 BV·h^-1(BV:body volume); 70 % alcohol was selected as elution agent. Conclusions The technology is economic and applicable. The yield of total saponin is no less than 75 %.
