摘  要:目的建立反相高效液相色谱法测定人血浆中克林霉素浓度的方法。方法采用反相高效液相色谱法,NUCLEODUR C18柱(150mm×4.6mm,5μm),流动相为乙腈-四氢呋喃-磷酸缓冲液(27:1:72),流速:1mL·min^-1,测定波长:204nm。结果克林霉素最低定量浓度为0.05μg·mL^-1,线性范围为0.05-16μg·mL^-1(r=0.9983),方法回收率均〉95%。日内、日间精密度(RSD)〈10%。结论该方法快速、简便、无杂质干扰,适用于克林霉素的体内药物浓度测定及药动学研究。Objective To establish a reversed phase high-performance liquid chromatography method to determine clindamycin in the human plasma. Methods RP-HPLC was used with NUCLEODUR C18 column (150 mm×4.6 mm, 5 μm), and acetonitrile-furan-H3PO4 buffer (27 : 1 : 72) was used as the mobile phase. The detection wavelength was 204 nm and the flow rate was 1 mL · min^-1. Results The calibration curve was linear within 0.05 - 16μg · mL^-1 (r=0. 998 3). The method recoveries were higher than 95%. The RSD values of intra-day and inter-day were lower than 10%. Conclusion The method is accurate, sensitive, specific and reliable for clindamycin. It may be used to study the pharmacokinetics of clindamycin.
