摘 要:实验研究了从南极采集的冰藻 Chlamydomonas sp.L4经强 UV-B 辐射胁迫后的生物学效应及逆境适应性。研究显示,经过强 UV-B 胁迫,冰藻 L4细胞内迅速产生大量的活性氧,丙二醛含量升高;同时,SOD 活性快速提高,使活性氧和丙二醛含量维持在细胞容许范围之内。膜脂肪酸成分分析发现不饱和指数增加了近4倍,膜流动性增加,从而维持膜的正常功能。光学显微镜、扫描和透射电镜观察表明,UV-B 辐射使细胞体积显著增大,黏性多糖分泌增多,细胞壁增厚,未知黑色颗粒密度增加,L4可能通过上述结构变化把 UV-B 屏蔽在胞外。脂肪颗粒的增加是细胞在逆境生存的能量保障。此外,细胞中类囊体片层、线粒体和细胞核等结构基本没有变化,维持细胞基本的代谢和能量供应。The effect of ultraviolet B (UV-B) radiation on an ice microalga sample collected from the Antarctica and its adversity adaptation were experimentally studied. The study showed that under stress of strong UV-B radiation, much active oxygen generated promptly in the sample cells, which led to the accumulation of MDA. In addition, the activity of SOD increased rapidly to maintain cells in a normal metabolic status. The analysis of fatty acid composition showed that the index of unsaturated fatty acids (IUFA) in cytoplasm membrane increased by nearly 4 times, and the fluidity of membrane accelerated accordingly. The results of optical, scanning and transmission electron microscopy showed the volume of the sample increased obviously under UV-B stress. The secretion of extmcellular substances and increasing depth of cell wall shielded UV-B radiation rightly. Fat granules increased and supplied energy to cells in adversity. Additionally, the structure of thylakoid, mitochondria and cell nucleus changed slightly under UV-B radiation, which maintained the fundamental metabolism of organisms.
