摘  要:目的比较低血清浓度(0.4%)和正常血清浓度(10%)培养条件对TGF-β1刺激成纤维细胞增殖的影响。方法原代大鼠皮肤成纤维细胞分别采用0.4%和10%血清浓度培养,加TGF-β1刺激细胞后,分别用BrdU ELISA法检测细胞增殖情况,噻唑蓝(MTT)比色法制作细胞生长曲线。结果0.4%和10%血清浓度下,250pg/ml TGF-β1均促进成纤维细胞增殖(P<0.01),并促使生长曲线的峰值提前和增加。而25ng/ml TGF-β1均抑制成纤维细胞增殖(P<0.05),并导致生长曲线峰值的滞后和降低。结论血清浓度对大小剂量TGF-β1刺激成纤维细胞增殖变化无明显影响。Objective To compare the effects between low - concentrated FBS ( Fetal Bovine Serum) ( 0. 4 % ) and high - concentrated FBS ( 10% ) on fibroblast proliferation induced by TGF - β1 ( transforming growth factor beta - 1 ). Methods Primary dermal fibroblasts of rats were cultured by low concentration FBS (0.4%) and high concentration FBS (10%) medium respectively; after TGF- β1 was added, MTT assay was applied to detect the fibroblast proliferation and BrdU ELISA method was applied to made out cell growth currve. Results In both low concentrated FBS {0.4%) and high concentrated.FBS (10%) condition, 250pg/ml TGF- β1 promoted cell proliferalion ( P 〈 0.01), the peak of the cell growth curve appeared earlier and its values increased; 25ng/ml TGF- β1 inhibited cell proliferation ( P 〈 0.05 ), and led to a later appearance of the peak of the cell growth curve and a decrease of its values. Conclusions The FBS concentration has no significant influence on the fibroblast proliferation induced by high or low dose TGF- β1.
