摘  要:目的探讨主动外排泵在鲍曼不动杆菌诱导突变株对氟喹诺酮类药物敏感性的影响。方法用环丙沙星诱导鲍曼不动杆菌筛选突变株。采用琼脂二倍稀释法检测泵抑制剂羰基氰氯苯腙(carbonylcyanide-m-chlorophenyl-hydrazone,CCCP)或利舍平存在时鲍曼不动杆菌诱导突变株对环丙沙星、左氧氟沙星和加替沙星敏感性的变化。结果CCCP和利舍平能影响部分鲍曼不动杆菌突变株对3种氟喹诺酮的最低抑菌浓度(minimum inhibition concentration,MIC),使MIC降至原值的1/4以下,泵阳性株在对3种氟喹诺酮均敏感的突变株中即存在。结论主动外排泵表达增强可能是鲍曼不动杆菌突变株MIC值升高的原因之一。Objective To investigate the role of efflux pump of Acinetobacter baurnannii mutant strains against fluoroquinolones. Methods The mutant strains of Acinetobacter baumannii separated from clinical sample were selected by ciprofloxacin. The antibiotic activities of ciprofloxacin, levofloxacin and gatifloxacin were detected by 2-fold agar dilution method with or without efflux pump inhibitors, carbonylcyanide-mchlorophenyl-hydrazone (CCCP) or reserpine. Results CCCP and reserpine could reduce the minimum inhibition concentration (MIC) to below 1/4 of former in some of the mutant strains against fluoroquinolones. Positive efflux pump strains existed in mutant strains which were susceptible to them. Conclusion The excessive expression of efflux pump was related to the higher MIC in mutant strains of Acinetobacter baurnannii.
