摘 要:目的比较5种不同化合物对病原菌金属β-内酰胺酶(MBL)的抑制作用,建立一种检测MBL的简便方法。方法在亚胺培南(IMP)纸片上滴加不同浓度的抑制剂,观察各自的抑菌环大小和清晰程度(Hodge试验);以IMP作为指示药物,在相邻的纸片上加抑制剂,即双纸片协同试验(DDSTs),再对抑制剂浓度和两者间的距离进行条件优化,选择出检测MBL的最佳方法。结果Hodge试验和DDSTs中均以巯基乙醇对MBL的抑制作用最强,将其作1∶15稀释后,巯基乙醇自身的抑菌环消失,与IMP纸片相距3 mm时,可清晰而准确地检测出MBL。结论巯基乙醇和IMP双纸片协同试验可优化MBL的检测。OBJECTIVE To establish a simpler method for screening bacterial metallo-β-lactamase (MBL) by comparing Hodge test and double-disk synergy tests (DDSTs), and using 5 inhibitors such as sodium citrate, EDTANa2, EDTAK2, sodium dimercaptopropane sulfonate and mercaptoethanol. METHODS In Hodge test, 5 inhibitors of different concentration were added to the imipenem(IMP) disks, the sizes of the inhibition zones were compared. Disk containing IMP and disk containing a metallo-β-lactamase inhibitor were used in DDSTs. The optimal inhibitors concentration and edge-to-edge distance between the two disks were selected. The performances of the Hodge test and the DDSTs were also compared. RESULTS Among the metallo-β-lactamase inhibitors used in this study, 1 : 15 diluted mercaptoethanol gave the most reproducible and the clearest results when a filter disk containing mereaptoethanol was placed near the IMP disk with the edge distance of 3 mm. CONCLUSIONS Mercaptoethanol-IMP DDSTs are simple, convenient and sensitive methods for screening MBL.
