摘 要:目的观察山茱萸环烯醚萜苷(comel iridoid glycoside,CIG)对血栓形成和凝血功能的影响。方法大鼠灌胃给予不同剂量的CIG(20,60,180 mg·kg^(-1)),连续7 d,于末次给药后1h建立动-静脉旁路形成模型,测量血栓干重和湿重;并进行体外血栓形成实验,测定血栓长度、干重和湿重。取新西兰兔抗凝血,分离血浆,加入不同剂量CIG(10,20,40 mg·mL^(-1)),测定凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)和凝血酶时间(TT)。结果CIG各剂量能明显抑制动-静脉旁路实验大鼠血栓形成,减轻血栓干重和湿重;缩短体外血栓实验大鼠血栓长度,减轻其干重和湿重。CIG可明显延长TT,中、大剂量组与生理盐水组比较有显著性差异(P<0.01);延长PT,大剂量组与生理盐水组比较有显著性差异(P<0.01);延长APTT,各剂量组与生理盐水组比较均有显著性差异(P<0.01)。结论CIG具有抑制血栓形成和抗凝血的作用,为CIG用于防治缺血性血管疾病提供实验依据。Objective To investigate the effect of cornel iridoid glycoside (CIG) on thrombosis and blood coagulation. Methods CIG at different doses (20, 60, 180 mg· kg-1)was intragastrically administered to rats for 7days, and the thrombosis model in vivo was established through setting up arteriovenous side road one hour after last administration in rats. The in - vitro thrombosis test was carried out by using a machine in which a revolving ring was installed. The weight and length of thrombi were measured in the two tests. Blood coagulation test in vitro was conducted in rabbits, and prothrombin time (PT), activated partial thromboplastin time (APTT) and thrombin time (TT) were measured after different doses of CIG(10, 20, 40 mg· mL-1) were added in the plasma. Results CIG at the doses of 20, 60 and 180 mg/kg significantly decreased both wet weight and dry weight of thrombi in arteriovenous side road thrombosis test, and obviously decreased the thrombi length, wet weight and dry weight of thrombi in vitro. Compared with the saline group, TT was prolonged in CIG groups at the doses of 20 and 40 mg/kg, PT was prolonged in 40 mg/kg CIG group, and APTT was prolonged in CIG groups at different groups ( P 〈 0.01 ). Conclusion CIG possesses the effect of inhibiting thrombosis and blood coagulation, which will provide evidence for the prevention and treatment of ischemic vascular diseases.
