摘 要:目的观察丹参酮IIA对大鼠系膜细胞增殖及对人TGF-β1基因启动子活性的作用。方法应用MTT法观察丹参酮IIA对大鼠系膜细胞增殖的作用;将不同长度的人TGF-β1基因启动子片段与氯霉素乙酰基转移酶(CAT)报告基因组成重组体phTGF 2.14,采用脂质体法转染至大鼠系膜细胞中,分别加入不同浓度的丹参酮IIA(1、5、10 mg/L)进行干预,用ELISA方法检测报告基因CAT的活性。结果10 mg/L的丹参酮IIA对大鼠肾小球系膜细胞增殖有明显的抑制作用,与对照组比较有明显差异(P<0.01)。丹参酮IIA浓度为1、5 mg/L时对重组体phTGF 2.14的表达活性无影响,当增大丹参酮IIA浓度为10 mg/L时对重组体phTGF2.14的表达活性有抑制作用,与对照组比较有显著差异(P<0.01)。结论丹参酮IIA能够抑制系膜细胞增殖,当浓度为10 mg/L时对人TGF-β1基因启动子活性也具有抑制作用。Objective To observe the effect of tanshinone ⅡA on the (RMC) and the gene promoter activities of human TGF-β1. Methods proliferation of rat mesangial cell The effect of tanshinone ⅡA on the proliferation of RMC was observed by using methyl thiazolyl tetrazolium (MTF) test. The segments of gene promoters of human TGF-β1 in different lengths and reporter gene of chloramphenicol acetyhransferase (CAT) were constructed to the recombination plasmid phTGF 2. 14, which was transformed to rat RMC by liposome method and then intervened by tanshinone ⅡA in different concentrations (1 mg/L, 5 mg/L and 10 mg/L). The activities of CAT reporter gene were detected by using enzyme-linked immunosorbent assay (ELISA). Results Tanshinone ⅡA ( 10 mg/L) had remarkable inhibitory effect on the proliferation of RMC in the tanshinone ⅡA groups with a significant difference compared with that in the control group (P 〈0.01 ). In tanshinone ⅡA groups when the concentration of tanshinone ⅡA was 1 mg/L or 5 mg/L the expression activities of the recombination plasmid phTGF 2.14 had no changes. When the concentration of tanshinone ⅡA was 10 mg/L the expression activities of the recombination plasmid phTGF 2.14 were inhibited, and had a significant difference compared with those in the control group (P 〈 0. 01 ). Conclusion Tanshinone ⅡA has inhibitory effect on the proliferation of RMC. When its concentration is 10 mg/L, tanshinone ⅡA can also inhibit the gene promoter activities of human TGF-β1.
